The game is mechanically the same as the first Hole in One; the player needs to hit the golf balls as Monkey and Mandrill throw them. However, this game has Monkey throw the golf balls with less delay in between, and will throw many of them in a row, requiring the player to hit them faster. Mandrill's offbeat throws are much more frequent as well, at one point throwing two in a row.

Hole in One 2 (ホールインワン レディー?, Hole in One Lady), known in Rhythm Heaven Fever as Hole in One 2 (ホールインワン2?, Hōru in Wan 2), is the 3rd Rhythm Game of Stage 9 (43rd overall) in Rhythm Heaven Fever and 3rd of Right-Hand Tower (83rd overall) in Rhythm Heaven Megamix.
