In this game, the player controls The Wandering Samurai who is once again slicing Demons, who have kidnapped a woman in the village.

Gameplay
During the game, the Demons spawn from all corners of the screen, coming out of thin air or from the water, with the samurai slicing them at the right time. In the middle of the game, he boards a skateboard, slicing demons on the move. Occasionally, a big demon appears, who the samurai must first block before slicing. At the end, the samurai faces the purple large demon who had kidnapped the woman.

The Wandering Samurai must slice those dark creatures to rescue the princess being kidnapped by their leader. During the game, he will jump on a skateboard, which will make it move quicker to rescue her. Press A or D-Pad button to slice a small demon that pops up from four sides and even one from the river. When you hear a scything tune, that means there is a big demon coming for you to attack you, hold B button to counter its sword attack and then release it to slice a big demon in right time.

Trivia
*In the Korean version of Rhythm Heaven Megamix, the Wandering Samurai and the woman have their traditional Japanese clothing altered to resemble Korean traditional clothing.
*This Rhythm Game is designed to resemble video games from the 8-bit and 16-bit era. This extends to the title using the word "Super", as many games did back in the day. The prologue having "2015 MADE IN KYOTO" beneath the game's logo and options for "1 PLAYER" and "2 PLAYERS" references how the earliest games from the Nintendo Entertainment System and arcade games from around that time would display their copyright and player options, as well as the year in which Rhythm Tengoku: The Best+ was released, and the location of Nintendo's headquarters, Kyoto, Japan.

*The scream the lady makes upon being kidnapped is reused from Tap Trial when Girl misses a jump.
* This rhythm game has a 16-bit visual style.
*Like in Iai Giri and Iai Giri Gaiden, the Japanese title of this game uses a nearly obsolete Japanese kana, wikipedia:Wi (kana), which is rarely used in modern Japanese.
* The words "2015 MADE IN KYOTO" on the bottom of the title screen are referencing the year that the Japanese version of Megamix was made, and the location of Nintendo's headquarters, Kyoto, Japan.
*This game appears in the "Wario...Where?" Challenge Train course. Here, The Wandering Samurai is replaced with 9-Volt and 18-Volt, who must rescue Sal Out instead of a lady.
* The girl from tap trial has the same scream as princess when she fails her jump
Development History
* Just like Samurai Slice for the GBA, it has an old letter (ゐ) which is rarely used in current Japanese writing.
Super Samurai Slice/Development
* This game appears in the "Wario…Where?" Challenge Train. In this game, you play as 18-Volt piggybacking 9-Volt in place of the Wandering Samurai, who are slicing bad ghosts while saving Sal Out, who replaces a townfolk maiden, kidnapped by these ghosts.
In Other Languages
* This game has identical controls to Fork Lifter, with the exception of the big demon.
