In this game, the player portrays a Student, under the teaching of DJ Yellow, a jolly fellow with the signature phrase "Scratch-o! Hey!". To play, after DJ Yellow says "Break, come on! Ooh!" or "And stop! Ooh!", the player must Tap and hold on the "Ooh!". Then, when DJ Yellow says "Scratch-o! Hey!", the player must flick on the "Hey!".
This game is about the titular DJ School, where DJ Yellow teaches his Student (controlled by the player) how to look like a DJ. DJ Yellow gives cues like "Break, come on! Ooh!", "And stop! Ooh!", and "Scratch-o, hey!". When he says "Ooh", the player needs to tap and hold to stop the record, and when he says "Hey", the player needs to flick to get it moving again.

*Perfect!/Ace!: The DJ routine will continue normally, and DJ Yellow will look at the Student with a very ecstatic look. Also, a white flash in the background will appear.

*This is one of the few games in the franchise to include a player input before the music starts. Others include Rap Women, Coin Toss, Remix 5 (Wii), and Figure Fighter 3.
*DJ Yellow and Student make a cameo appearance in Cheer Readers during one of the "OK, it's on!" cheers in Rhythm Heaven Fever and Rhythm Heaven Megamix.
*DJ Yellow and his student make a cameo appearance in Cheer Readers during one of the "OK, it's on!" cues.
**They later make another cameo appearance as the masks in Spaceball in Lush Remix in Rhythm Heaven Megamix.
*The Student makes a cameo appearance in the Try Again epilogue of Catch of the Day as a fishmonger.
*The Student makes a cameo appearance in the Try Again epilogue to Catch of the Day as a fishmonger in Rhythm Heaven Fever and Rhythm Heaven Megamix.
*DJ Yellow has two unused sprites; one where he is seen giving a thumbs up, and another looking cross; the thumbs up sprite was probably used when getting a perfect hit in-game, and the cross sprite might have been meant to be used in an epilogue.
*Sliding the stylus around after stopping the record produces scratching noises. This does not affect gameplay in any way.

*This, along with Space Dance, Night Walk, Hole in One, Tambourine, See-Saw, and Fruit Basket are the only games to have names that are functionally identical to their Japanese name.
